- I forgot to say that the tagged tubulin incorporates very well in the spindle, here after a 4-hour induction (green, magenta is axoneme + transition zone). However, we could not see a defined polarity, in contrast to flagellar or cortical microtubules.

- Thanks Martin, actually we started these experiments with @keithrmatthews.bsky.social and Azadeh Bagherzadeh in Manchester when we were still in Keith's lab! Although we could express different versions of tagged tubulin, they all failed miserably to be incorporated into microtubules.

- I fully agree! This is such a conceptual and clever paper and has been a source of inspiration for me and many colleagues working on cilia and flagella!

- We propose a model where the new flagellum grows in a linear manner while the old one is locked but shrinks slightly, hence requiring unlocking and compensation after cell division, leading to such multicoloured flagella following induction/deinduction/reinduction of tagged tubulin 😉
- Second, the old flagellum is indeed locked during assembly of the new flagellum, as we proposed previously www.sciencedirect.com/science/arti... rupress.org/jcb/article/...
- Surprise! Some incorporation takes place in old flagella inherited after cell division!
- In contrast to Chlamydomonas, flagellum growth rate is linear
- Great work showing that cilia are worth even more than motility! 👏👏👏

- By FIB-SEM, Aline found multiple densities resembling IFT but thinner and shorter than IFT trains encountered on external portions of flagella. Strikingly, they appear to fuse (or split?) and spread along distinct DMT as if they were able to switch from one DMT to another!
- To demonstrate the molecular identity of these densities, Aline used #UExM coupled to STED to reveal that they indeed contain IFT material that displays the same positioning as the densities detected by FIB-SEM
- dSTORM reveals that IFT proteins are found around all 9 doublet microtubules
